External root resorption (RRE) is the loss of dentine and cement, is of multifactorial origin, includes mechanical and biological factors. The objective of this research was to evaluate biological and orthodontic factors on the RRE induced by orthodontic treatment. A quantitative longitudinal study with a prospective design that included 30 patients treated orthodontically with Roth and MBT techniques (McLaughlin, Bennett, Trevisi) was performed. Participants were given initial periapical radiographs (T0) before starting treatment and at 6 months (T1) of having started orthodontic treatment, to identify if RRE was present. Blood samples were taken from which DNA was extracted and SNP IL6 rs1800796 (-572G> C) was genotyped by PCR and digestion of the product with the enzyme BsrBI. In a profile radiograph, the type of malocclusion, degree of initial dental inclinations, and in models the degree of dental crowding were evaluated. The statistical analysis was carried out with the use of the SPSS V 22 program, tables and graphs were made to describe the absolute and relative frequencies in the univariate analysis. The Chi2 test was used for the bivariate analysis. A multivariate correspondence analysis was used to compare all the variables with the IL6 genotype rs1800796 (-572G> C). Variables such as moderate dental crowding were associated with RRE (p-value 0.026), there is also higher RRE in patients with class II malocclusion (p-value 0.029). There was an association between the IL6 polymorphism rs1800796 (-572G> C) with the RRE in the patients at the beginning of the orthodontic treatment. Moderate dental crowding and class II malocclusion were associated with the RRE, there was an association between the polymorphism rs1800796 (-572G> C) of the IL6 gene with the RRE in the patients at the beginning of the orthodontic treatment
