The success of endodontic treatment has been associated with the timely sealing of the endodontic filling material and the absence of microfiltration in the restoration. However, it is widely known that there are situations that require this type of treatment to be performed in more than one appointment, either due to the complexity of the procedure or the time allotted to the appointment for its completion. For this reason, the use of temporary filling materials is used to protect the root canal system once its preparation has begun until the final filling is achieved; thus avoiding the entry of pathogenic microorganisms into the duct system and a probable reinfection that leads to treatment failure. The objective of this research was to determine which was the separator with the lowest degree of microfiltration between the cotton ball and the PTFE tape in the temporary seal in endodontics. A comparative experimental study was carried out in which 30 healthy single-rooted premolars were used, donated by patients with an indication for extraction for orthodontic purposes; which were stored in 1% chloramine T for a period no longer than six months, the teeth were subjected to conventional endodontics, they were randomly divided into three groups: GI n = 10 temporary seal with cotton, G II n = 10 temporary seal with PTFE tape, G III n = 10 control group without separator. Subsequently, the specimens were divided into two groups and subjected to a thermocycler process for 50 cycles (7.5 hours) and 100 cycles (15 hours), simulating 15 and 30 days in the oral cavity, with a previous thermal bath for 30 seconds at temperatures of 5 and 55 ° C. They were sequentially immersed in a 4% methylene blue solution for 24 hours, after which, with a low speed diamond disc, each tooth was fragmented into 2 sections and subsequently taken for exploration with a stereoscopic microscope; Through the use of photographs, the degree of penetration or microfiltration was measured. It was evidenced that the control group, that is, the specimens that did not use separating material between the temporary cement and the plugged canals presented greater leakage (median = 11.5mm IQR 3.2-21.4), followed by the group with the separator of cotton (4.6m IQR 3.5-11.4) and finally the PTFE tape which presented the least leakage (2.6mm IQR 1.2-3.9). The use of a spacer such as 3 PTFE (Teflon) tape contributes to significantly reduce leakage during temporary filling of endodontic treatment.
