Assess the levels of SP, CGRP, its NK-1 and CGRP-R receptors in the periodontal ligament of the lower premolars after root canal preparation using the Reciproc blue®, M-Two® and Plex V® systems. Fifty periodontal ligament samples were obtained from lower premolars extracted for orthodontic reasons, which were randomly distributed to five groups: a negative control or without instrumentation, a positive control prepared with manual files, and 3 preparation systems (Reciproc Blue® , M-Two® and Plex V®) (n = 10). The tissues were stored at -80 ° C, then thawed in a cold bath and refrozen with their respective thaws to disintegrate the tissue. Three sonication cycles were performed until the samples were homogenized, then centrifuged and the supernatant was collected to detect the markers to be studied. The samples were processed for the ELISA test using the ELISA-sandwich principle. For each analyzed protein, the mean, median and standard deviation were calculated in each group. The ANOVA and Kruskal-Wallis tests were performed according to their parametric or non-parametric behavior. Finally, differences between groups were explored by means of a Tukey test in normality or U Mann-Whitney, if they were not normal.
